- 1,4 mg/m² i.v. (max. 2 mg Einzeldosis!)

Indikationen (الاستطبابات)

- ALL, Lymphome, Wilms-Tumor, Neuroblastom

Kontraindikationen (موانع الاستخدام)

- NIEMALS intrathekal (tödlich!)

Prüfung الامتحان · Exam

Merksatz: (جملة للحفظ) Vincristin NIEMALS intrathekal → sofortiger Tod! Max. 2 mg Einzeldosis (unabhängig von KOF).

Nebenwirkungen (الآثار الجانبية)

- Neurotoxizität (Haupttoxizität!): periphere Neuropathie, autonome Dysfunktion, Obstipation

Interaktionen (التفاعلات)

- CYP3A4-Inhibitoren (Azol-Antimykotika): Vincristin-Spiegel ↑ → Neurotoxizität ↑
- Mitomycin C: pulmonale Toxizität ↑ [VERIFY]
- Phenytoin: Spiegel ↓

Wirkmechanismus (آلية التأثير)

Vincaalkaloid → bindet Tubulin-Dimere → hemmt Tubulin-Polymerisation → Mitosespindelzerstörung → Zellzyklus-Arrest in Metaphase → Apoptose.

Monitoring (المراقبة)

- Neurologische Untersuchung (Reflexe, Sensibilität, Parästhesien)
- Stuhlgang (autonome Neuropathie → paralytischer Ileus!)
- BB (Myelosuppression gering)
- Max. Einzeldosis 2 mg einhalten!
